A sustainable alternative to deep-freezing for plasma preservation.

Objective

To compare the quality and functionality of plasma samples stabilized and stored at room temperature vs. frozen at -80°C, in the context of a prospective collection of biobank samples.

Materials and Methods

Plasma obtained from peripheral blood in EDTA and citrate

Procedure

The collected samples are evaluated according to established quality control (QC) criteria before being divided into four fractions: two for lyophilization and two for freezing at -80°C. Aliquots of 500 µL are prepared for lyophilization, which is performed on the same day and concurrently with the freezing of parallel aliquots. All lyophilized samples are stored at room temperature.

The quality of fresh samples will be analyzed prior to lyophilization, while the stability of both frozen and lyophilized samples will be evaluated after 1 and 6 months of storage.

Quality Assurance

Electrophoresis followed by Coomassie staining

ELISA

Quality control will include ELISA test and polyacrylamide gel electrophoresis followed by Coomassie staining to analyze the protein profile. The signal intensity of the predominant bands obtained in the gels will be quantified, allowing an accurate assessment of protein integrity and stability under each storage condition.
